If I recall my history classes correctly, many nuns were in fact trained in self defense, and those that came from wealthy families often already had professional training in grappling and knives. Most convents and seminaries were forbidden to use weapons of war though, so tools were often taught. Staves, hammers, even machetes and (hunting) spears, as those were seen as tools before weapons. Nuns were, far more often than not, much more capable of defending themselves than media and modern sensibilities allow shown. Even today, Jujitsu and Judo are popular among those that take the cloth. Source, the teachers of my theology classes back in high school. The Nuns and Priests could often be found training together after hours.

Some nuns might have had some martial training. Coming from nobility, they might have received some formal training in defense. And women from the more practical social classes still had to know where to kick or poke with a pointy implement to dissuade assaults.
Skill in self defense was useful even then.
