For context on the relationship between Lancelot and Arthur bc the show doesn't explain, Lancelot (who was Arthur's greatest friend and best swordsman in Camelot) fell in love with Guinevere, Arthur's wife. They had an affair and long story short, Arthur found out, Lancelot fled, and Guinevere was sentenced to death. Lancelot returned to save her and killed a ton of the knights of the round table which sparked a conflict that divides the knights and ultimately leads to Camelot's downfall. Guinevere is exiled and lives as a nun, and Lancelot becomes a hermit. That's why in the show, Lancelot is saying he wished Arthur had judged him more harshly, since he spent the remainder of his life eaten up by the guilt of what he caused.

He also has his own spinoff show, Lord El-Melloi's Case Files, which isn't about the Grail War. It's more of a detective show where he takes on cases.

We're constantly seeing Gilgamesh downing bottles of it, and when he first starts tempting Kirei, Kirei refuses to drink any. He even calls Gilgamesh blasphemous and sinful, because he's still trying to cling to the ideal of being a moral man. However, once he gives in to Gilgamesh and starts toying with Kariya in episode 21, he finally drinks, and when asked how it is, he responds by saying he'd love to sample it again, marking the completion of his "fall" and how he's finally accepted that he derives pleasure from the misfortune of others.
