That's because, just as Glynda doesn't respect Jauncifer, he doesn't respect her either. Sepper isn't just a badass, she knows her stuff and is helping them refine their combat capabilities. Glynda, who doesn't even weild a true weapon, taught the combat class just by making students constantly fight each other while giving manifestly bad advice. Now, that's really because RWBY's writers are terrible at their jobs, but I've spun that into the woman being a damn good Huntress, but an absolutely terrible teacher, and thus Jaune treats her like a more experienced Huntress, not a proper instructor. So, when she demands, to him, unearned respect, he needles her instead.

I'm guessing it's his Dragon-ness making him disregard proprietary when addressing Glynda, an older female teacher, who clearly strives to create a divide between her personal and professional life, by her given name. He doesn't do that to Sepper. I mean, he can see her as his contemporary, but if Glynda isn't comfortable with doing the same, shouldn't he be respectful of that? It feels like such a weird, pointlessly needling thing to do.
